摘 要:采用Plackett-Burman试验设计筛选显著影响多糖产量的培养条件,然后用最陡爬坡路径逼近最大响应区域,最后通过Box-Behnken设计及响应面分析确定主要影响因素的最佳值。结果表明,多糖发酵培养条件为:初始p H值6.5、摇床转速209 r/min、装液量48 m L/250 m L、温度31℃、接种量10%、种龄90 h、培养时间180 h。在此最佳条件下,黄伞发酵胞外多糖产量可达7.44 g/L,比优化前提高了近2倍。利用响应面分析法优化黄伞胞外多糖发酵培养条件是可靠的,具有实际的应用价值。Plackett-Burman design was used to select the fermentation conditions of significant impact on the polysaccharide yield, and then the path of steepest ascent was adopted to approach the optimal region. Finally the optimum levels of the main influencing factors were determined by Box-Behnken design and response surface analysis. The results showed that the optimum fermentation conditions of exopolysaceharide were initial pH 6.5, shaking speed 209 r/min, liquid volume 48 ml/250 ml, temperature 31℃, inoculum 10%, inoculum age 90 h and culture time 180 h. Under the optimum conditions, the yield of Pholiota adipose exopolysaccharide could reach to 7.44 g/L, which was about 2 times than that of before the op- timization. It was reliable that the fermentation conditions of P. adipose exopolysaccharide were optimized by response surface analysis, which had practical application value.
