摘  要:本文主要考察葡萄糖对毕赤酵母生长和组成型表达α-葡聚糖酶的影响。采用单因素实验法考察初始发酵培养基中葡萄糖浓度、补料阶段葡萄糖残留、通气量等对α-葡聚糖酶产量的影响。结果表明发酵培养基中初始葡萄糖浓度50g/L最佳,提高初始葡萄糖浓度,毕赤酵母生长和α-葡聚糖酶产量都明显受到影响。补料过程葡萄糖残留越小越有利于酶的表达,最理想的控制应该是葡萄糖残留几乎为零,但又保证能满足菌体的生长和表达需要。提高通气量有利于增加α-葡聚糖酶的产量,发酵时间66h为宜。在优化条件下,α-葡聚糖酶酶活达16537U,为利用葡萄糖生产α-葡聚糖酶打下基础。This paper mainly to investigate the effects of glucose on Piehia pastoris growth and constitutive expression of Dextranase. Examined the initial concentration of glucose in fermentation medium, glucose concentration in fed-batch, ventilation and other effects on constitutive expression of Dextranase. The results showed that: the best initial glucose concentration in fermentation medium is 50g/L, when increase the initial concentration of glucose, the growth and Dextranase expression is appeared to be markedly reduced. Glucose concentration between 0 to 1 g/L in fed-batch, growth and Dextranase expression is the best, the ideal control should be glucose concentration is almost zero, but ensure that can meet the requirements of the growth of Pichia pastoris and expression of Dextranase. At the same time,we found that improve the ventilation is helpful to the expression of Dextranase and suitable fermentation time is 66h. Under this condition,Dextranase activity reached about 16537U.
