摘  要:目的探讨法呢醇X受体(FXR)、瘦素(Lep)及髓过氧化物酶(MPO)在胆管癌组织中的表达变化及意义并分析它们间的关系。方法采用免疫组织化学染色及Real time-PCR法检测胆管癌组、癌旁无癌胆管组和正常胆管组中FXR、Lep和MPO的表达。结果 FXR、Lep和MPO在胆管癌组、癌旁无癌胆管组和正常胆管组中均有表达。FXR在癌旁无癌胆管组织和正常胆管组织中的阳性表达率显著高于癌组织(P<0.05),而在癌旁无癌胆管组织与正常胆管组织中,无显著性差异(P>0.05)。Lep和MPO在癌旁无癌胆管组织和正常胆管组织中的阳性表达率显著低于胆管癌组织(P<0.05),而在癌旁无癌胆管组织与正常胆管组织中,无显著性差异(P>0.05)。FXR蛋白与Lep蛋白之间呈线性相关关系(r=-0.738,P<0.05),而FXR、Lep与MPO无相关性(P>0.05)。结论 FXR、Lep与MPO可能通过调节脂质代谢平衡及免疫应答等机制共同参与胆管癌的发生和发展,进一步的研究有可能为胆管癌的诊断和治疗提供新的途径。Objective To investigate the farnesoid X receptor (FXR), leptin (LEP) expression in bile duct cancer and to analyze the relationship between them. Methods and myeloperoxidase (MPO) The expressions of FXR, Lep and MPO in the bile duct cancer group, adjacent non-cancer group and the normal bile duct group were detected by im- munohistochemical staining and Real time PCR method. Results FXR, Lep and MPO in the bile duct cancer group, adjacent non-cancer group and the normal bile duct group were expressed, the positive expression rate of FXR in the ad- jacent non tumor margin and normal bile duct tissues were significantly higher than bile duct carver tissue (P 〈 0.05) , while in the adjacent non tumor margin and normal bile duct tissues, there was no significant difference (P 〉 0.05). The rate positive expression of Lep and MPO in the adjacent non tumor margin and normal bile duct tissues was signifi- cantly lower than bile duct cancer tissue (P 〈 0.05) , and in the adjacent non tumor margin and normal bile duct tis- sues, there was no significant difference ( P 〉 0.05 ). There was linear correlation between FXR and Lep ( r = - 0. 738, P 〈 0.05 ). While there is no correlation between FXR, Lep and MPO (P 〉 0.05). Conclusion FXR, LEP and MPO can regulate lipid metabolism balance and immune response mechanisms involved in the occurrence and development of bile duct carcinoma,further research may provide a new way for the diagnosis and treatment of bile duct cancer.
