摘 要:以正常人肝细胞L02为实验对象,采用乙醇诱导建立酒精性肝脏损伤的细胞模型,并以MTT法筛选出毛木耳蛋白的作用浓度范围。通过检测细胞内甘油三酯(TG),活性氧(ROS)含量以及胞外谷丙转氨酶(ALT)和谷草转氨酶(AST)含量来评价毛木耳蛋白保护酒精性肝脏损伤的功效。结果表明,毛木耳蛋白的作用浓度为30μg/m L时,其细胞活性较模型组提高42.8%,同时细胞内甘油三酯的含量明显降低,特征性指标明显改善,说明其可以恢复损伤细胞活性,改善酒精引发的脂肪堆积。通过游离活性氧检测发现细胞内活性氧的含量降低,说明毛木耳蛋白提取物可能是通过抗氧化途径发挥防护作用。Liver cells L02 were cultured with ethanol to establish the alcoholic hepatic injury model,and optimized the bioactive concentrations of Auricularia Polytricha protein extract by MTT assay.The protective effects of Auricularia Polytricha protein extract against hepatic injury were evaluated by the concentration detections of intracellular TG,ROS and extracellular ALT,AST.The results showed that Auricularia Polytricha protein extract could significantly increase cell viability with the ratio of 42.8% by the dose of 30 μg/m L.Compared with the model group,the content of TG and the enzyme activities of ALT and AST were significantly decreased in the extract-treated group.It indicated that Auricularia Polytricha protein extract repaired ethanol-induced cells damage and reduced fat accumulation in cells.In ROS assays,it implied that Auricularia Polytricha protein extract might play a protective role by means of antioxidant effect.
