摘 要:选择p H=7.2的磷酸缓冲液为浸提剂从富硒黄豆中提取可溶性硒蛋白,研究了液料比、提取温度、提取时间对黄豆蛋白提取率的影响。在单因素试验的基础上,根据Box-Benhnen的中心组合试验设计原理,采用三因素三水平的响应面分析法确定了富硒黄豆中硒蛋白提取的最佳工艺:液料比11∶1(m L/g);提取温度44℃;提取时间96 min;在此条件下,可溶性硒蛋白的提取率可达到76.03%。进一步研究表明:富硒黄豆中不仅含有硒蛋白,而且还含有大量的其他有机硒化合物。Soluble selenium-containing protein was extracted from selenium-enriched soybean by PBS(p H=7.2).The effects on protein extraction rate by the factors, such as liquid/ material ratio,temperature and extract time were studied. According to the single factor experiments,the optimum of extract process for selenium-enriched soybean protein was determined through Box-Benhnken central composite design and response surface methodology. The result showed that optimal condition of extraction were: liquid/material ratio 11 ∶ 1(m L/g);extraction temperature 44 ℃; extraction time 96 min.Under the optimized extraction conditions, the yield of soluble selenium-containing protein was 76.03 %. Further studies showed:selenium-enriched soybean not only had selenium-containing protein,but also had other selenium-containing compounds.
