摘 要:目的:研究大连紫海胆棘壳色素的体外免疫调节和抗炎活性。方法:以LPS(细菌脂多糖)为炎症诱导物,研究棘壳色素对RAW264.7细胞分泌多种炎症因子的影响。分别采用MTT法及中性红吞噬法研究棘壳色素对脾淋巴细胞增殖及腹腔巨噬细胞吞噬功能的影响。采用补体结合法考察棘壳色素对补体活性的影响。结果:棘壳色素可显著抑制LPS刺激后RAW264.7细胞合成分泌NO(作用质量浓度5~30μg/m L)、IL-6、TNF-α(作用质量浓度60μg/m L)、PGE2及6-KETO-PGF1α(作用质量浓度10~60μg/m L)的水平,但不影响IL-4的分泌水平。20,50μg/m L的棘壳色素均可促进脾淋巴细胞的增殖,30,60μg/m L的棘壳色素可显著增强腹腔巨噬细胞的吞噬活性,1~500μg/m L的棘壳色素还可显著抑制补体活性。结论:海胆棘壳色素可在功能性产品开发方面具有一定应用潜力。The anti-inflammation and immune-modulating activities of pigment from spines and shells of sea urchin(PSU) were studied. After the RAW264.7 cell was stimulated by LPS, the effect of PSU on some inflammation factors production was investigated. MTT and complement fixation test were carried out to detect the effect of PSU on lymphocytes proliferation and complement activities. Result: PSU could inhibit the production level of NO, IL-6, TNF-α,PGE2 and 6-KETO-PGF1α, but could not affect IL-4 level. PSU could stimulate spleen lymphocyte proliferation and abdominal macrophage phagocytosis activity. On the contrary, PSU demonstrated significant inhibition effect on complement activity. Conclusion: PSU was potential in functional products development, which might permit the comprehensive utilization of the spines and shells, by-product of sea urchin processing.
