摘  要:为了研究樟树籽油甘油二酯(DAG)的抑菌活性,以樟树籽油为原料,通过柱层析分离得到纯度为92.4%的DAG,采用滤纸片法、最小抑菌浓度(MIC)测定法、抑菌活力实验对4种细菌进行抑菌效果分析;通过菌液电导率、可溶性蛋白质与菌体核酸含量的测定和扫描电镜观察细菌表面形态变化,探究其抑菌机理。结果表明:DAG对G^+细菌和G^-细菌均有较好的抑制效果,抑菌活力均在0.60以上,抑菌时效3~48h。对G^+细菌和G^-细菌的MIC分别是2.0和4.0mg·mL^(-1)。加入DAG的菌液电导率升高、菌液可溶性蛋白质和菌体核酸增多,细菌表面形态发生改变,说明对细菌的抑制作用主要是通过破坏细菌细胞膜,从而使其内含物泄漏,菌体形态发生改变,抑制细菌生长。To research the antibacterial activity of the diglyceride(DAG)from camphor tree seed oil,used camphor tree seed oil as raw materials,the purity of DAG obtained is 92.4% by column chromatography separation.The antibacterial activity have been determinated on four types of bacteria by the filter method and minimal inhibitory concentration(MIC).The determination of bacteria liquid conductivity,soluble protein,nucleic acid content,and observation of bacterial surface morphology change by scanning electron microscope is to explore antibacterial mechanism of DAG.The results showed that DAG had a good inhibitory effect on G~+and G^-,antibacterial activity arrived over 0.60,the effect of antibacterial action up to 3~48h.The MIC of G~+and G^- were 2.0mg·mL^(-1) and 3.0mg·mL^(-1),respectively.After)adding DAG,bacteria liquid conductivity,soluble protein and nucleic acid of bacterium increased,and bacterial surface morphology changed.All the phenomena showed that the damaging of outer membranes can reach to inhibition action on the bacteria,thus cell contents leakage and bacteria morphology changes inhibit the growth of bacteria.
