摘  要:为了鉴定和了解实验室保存的1株溶原性鸡大肠杆菌的病原学特性,试验对其遗传进化和基因型等进行了分子特征鉴定。结果表明:鸡大肠杆菌Leco13的16S rRNA与大肠杆菌K12标准株16S rRNA部分序列的同源性达到了99%;用MLST分型特异引物对菌株的基因型进行鉴定,发现该菌株属于ST10型。同时参考其他文献中的噬菌体提取方法,对分离株进行了噬菌体的提取,成功提取噬菌体DNA后,用EcoRⅠ和HindⅢ进行酶切鉴定并获得了较为清晰的酶切图谱。To identify and understand the pathogenic characteristics of a strain of lysogenic avian Escherichia coli that preserved in a laboratory,its genetic evolution,genotype,and other molecular characteristics were identified in the test. The results showed that the homology between the16 S rRNA sequence of Escherichia coli Leco13 and partial 16 S rRNA sequence of the standard strain of Escherichia coli K12 reached 99%. The strain's genotype was identified by multilocus sequence typing( MLST) method using special primers,and it was found that the strain belonged to ST10. Meanwhile,the phage was extracted from the isolated strain according to the protocol of publishing literature. After the phage DNA was extracted successfully,the phage DNA was identified by EcoR Ⅰ and Hind Ⅲ digestion,and a clear restriction mapping was obtained.
