摘 要:目的采用响应面法对南极磷虾源磷脂的人参皂苷磷脂复合物制备工艺进行优化,并初步探讨其理化性质。方法以人参皂苷复合物的复合率为评估标准,采用单因素及响应面Box-Behnken设计考察各因素对复合物复合率的影响,优化制备工艺。利用扫描电镜观察磷脂复合物的固体粒子表面形态。结果最终影响磷脂复合物复合率的因素有3个:反应时间,人参皂苷浓度,投料比(人参皂苷:磷脂),最佳制备工艺为:以四氢呋喃作为反应溶剂,温度为40℃,反应时间为1.5h,投料比为1∶1.6,人参皂苷浓度为35mg·mL-1,在此条件下复合率可达98.08%.经3次验证实验平均复合率可达97.45%,实验值与预测值之间的相对误差为0.64%。结论响应面法对复合物制备工艺条件进行优化后,提高了复合物制备的复合率。Objective To optimize the preparation conditions of the Antarctic Krill phospholipid Ginsenoside phospholipid-complex by response surface methodology(RSM)and investigate its physicochemical properties.Methods The preparation conditions of Ginsenoside phospholipid-complex were optimized by means of single factor study and Box-Behnken design with the complex-rate of Ginsenoside and phospholipid as assessment criteria.The surface morphology of solid particles of Ginsenoside phospholipidcomplex was observed by scanning electron microscope(SEM).Results Three factors effected complexrate significantly including reaction time,concentration of Ginsenoside,ratio(Ginsenoside:phospholipid).The optimal preparation condition of phospholipid-complex:Using tetrahydrofuran as reaction solvent,the reaction temperature at 40 ℃,the reaction time was 1.5h,ratio was 1∶1.6,concentration of Ginsenoside was 35mg·mL-1.Under the optimal condition,the complex-rate was 98.08%.Three times validation experiments showed that the average complex-rate was 97.45%.Relative error between the actual value and predictive value was 0.64%.Conclusion After the optimization of preparation of phospholipid-complex by response surface methodology,the complex-rate was improved.
