摘  要:比较研究以脱盐乳清粉为主要原料,采用乳杆菌A-2进行发酵制备的代谢产物(LF),并初步探讨了LF抗氧化和抗肿瘤活性的功能。通过对发酵液的p H、总糖和Ca^(2+)含量的测定,筛选出最佳菌悬液浓度为1OD/mL;用ABTS法和ORAC法来测定和评价乳杆菌A-2发酵产物的抗氧化作用。在反应体系中,脱盐乳清粉和LF的ABTS清除率的IC50分别为34.29mg/mL和19.74mg/mL,LF的ABTS清除率的能力优于脱盐乳清粉;其相对ORAC值分别为75.46μmol Trolox/g和114.78μmol Trolox/g,LF清除AAPH的能力为脱盐乳清粉的1.5倍,结果显示LF具有一定抗氧化活性。采用MTT法检测LF对人舌癌细胞系Cal-27细胞增殖的影响,LF的IC50为19.1 mg/mL,且浓度在4~32 mg/mL的范围内,对人舌癌细胞系Cal-27细胞的抑制率呈剂量依赖性,与脱盐乳清粉组比较,差异具有显著性。In this study,the desalted whey powder was used as the principal raw material,using Lactobacillus A-2 for fermentation,to produce antioxidant activity and antitumor activity metabolite(LF). By measuring PH value of fermentation bacteria,total sugar and Ca(2+)concentration,the best concentration of the bacterial suspension is determined as 1OD /mL;the antioxidant activity of Lactobacillus A-2 whey fermentation product was evaluated by ABTS and ORAC. In the reaction system,the ABTS free radical scavenging activity IC50 of desalted whey powder and LF were 34.29mg/mL and 19.74mg/mL respectively,the ABTS free radical scavenging activity of LF is better than that of desalted whey powder;the ORAC assay of desalted whey powder and LF were 75.4 and 114.78μmol Trolox/g respectively. The results showed that LF had prominent antioxidant activity. Using MTT method to measure the influence of desalted whey powder and LF on Human tongue squamous carcinoma cells Cal-27 cell proliferation,the concentration of LF between 432 mg/mL had inhibition effect of Human tongue squamous carcinoma cells Cal-27 function on a dose-dependent manner,and the difference was significant compare with the desalted whey powder.
