摘  要:水解可大幅提高羊骨的营养价值和生物活性,水解程度越高,生物利用度越高,其抗氧化、促免疫等活性亦越强。本研究以碱性蛋白酶水解羊骨粉,探讨反映水解程度的水解度、短肽得率、氨基态氮含量、多肽生成量4个指标间的相关性,为水解效果评价提供最优指标。结果表明,单因素试验过程中,除温度外,各指标达到最大值时均具有相同的底物浓度(8%)、酶添加量(4%)、酶解时间(4h)和起始pH值(9),4个指标中,多肽生成量受各因素影响最大。相关性分析结果表明各指标之间均呈显著相关(P<0.01),与水解度的相关性(r值)依次为短肽得率(0.981)、氨基酸态氮(0.801)、多肽生成量(0.641),因此在优化羊骨酶解工艺时,水解度和短肽得率为优选指标,胶原肽经进一步分离纯化或直接干燥后,可广泛应用于饲料、食品、医药、化妆品等领域。Both the nutritional value and biological activities of sheep bone can be greatly improved by hydrolysis. The higher degree of hydrolysis,the higher bioavailability, and the higher antioxidant and immunomodulatory functions. To provide better evaluating indicators for hydrolysis,correlations of 4 indicators reflecting hydrolysis degree were analyzed using alcalase hydrolyzing sheep bone powder. Results of the single-factor tests suggested that except for temperature,the 4 indicators--degree of hydrolysis(DH), obtaining rate of short peptides, amino acid nitrogen, amount of polypeptides shared the same substrate concentration(8 %), enzyme supplementation(4 % ), reaction time(4 h) and the initiative p H (9) at the maximun. Polypeptide amount was affected obviously by all tested factors. Correlation analysis indicated that 4 indicators were significantly correlated with each other(P〈0.01). The correlation with the indicator of DH (r value) was obtaining rate of short peptides(0. 981) ,amino acid nitrogen(0. 801), polypeptides amount(0. 641). Therefore, DH and the obtaining rate of short peptides were the most important consideration during further optimization of the technology. The hydrolysate after further separation and purification, or dehydrated directly, could be widely used in the field of feed, food, medicine and cosmetic areas.
