摘 要:本研究采用浊度法,利用分光光度计测定菌液在560 nm波长的OD值,确定细菌浓度。通过测定酵母D7的生长曲线确定其生长周期,进而确定其培养时间;采用单因素试验确定出酵母菌生长的温度、转速、p H值的合适水平,正交试验L9(34)对生长条件进行优化,研究不同的温度、转速、p H值对优良废糖液酒精发酵酵母D7生长的影响。结果确定为16小时为酵母菌后续试验的培养时间;酵母菌的正交试验温度水平为30℃、32℃、34℃,转速水平为160 r/min、180 r/min、200 r/min,p H值水平为3、4、5;正交试验优化的最适生长条件为温度30℃、转速160 r/min、p H值为3。The vaccine OD value was measured by nephelometry at wave length of 560 nm to determine its cell concentration. The growth cycle and culture time was measured by growth curve of D7 strain. The suitable level of temperature,rotational speed and p H value of yeast growth was determined by single factor experiment. Orthogonal experiments L_9( 3~4) to optimize the growth conditions,the effects of p H value,temperature and rotational speed on the growth of D7 strain was studied. The results shows that the incubation time was 16 hours of follow-up experiments,level of orthogonal experiments temperature is 30℃,32℃,34℃,the speed level of 160 r / min,180 r / min,200 r /min,p H value level 3,4,5; Its optimum growth conditions were determined by orthogonal experiments as follows:temperature at 30℃,rotation speed 160 r / min,p H value of 3.
