摘 要:以匍枝根霉亚种点头根霉TP-02为原始菌株,利用ARTP诱变系统进行诱变处理.通过透明圈法进行初筛和摇瓶发酵进行复筛,获得一株遗传性状稳定的高产木聚糖酶诱变菌株TZD-01,木聚糖酶活力达到280.81U/mL,为原始菌株的2.7倍.通过光学显微镜和扫描电子显微镜观察诱变菌株与原始菌株微结构特征变化,ARTP诱变处理可引起菌株形态特征发生改变.诱变菌株的菌落形态不规则,颜色为黑褐色,比原始菌株偏深;菌丝直径变大,形态更饱满;孢子形态规则更接近圆形,诱变筛选菌株的生长速度比原始菌株更快.With Rhizopus stolonifer subspecies nod Rhizopus TP-02 as the starting strain,ARTP mutagenesis system was applied to mutagenic treatment.Primary screening by the method of transparent circle,then complex screening by shake flask fermentation,a stable xylanase producing strain TZD-01 was obtained.Xylanase activity reached 280.81U/mL,2.7times that of the original strain.The microstructure changes of the mutant strain and the original strain were observed by electron microscope and scanning electron microscope.ARTP mutation treatment can cause the changes of the morphological characteristics of the strain,irregular colony morphology of mutant strains,and the darker color than the original strain.After ARTP treatment,the mycelium diameter becomes larger,the shape is sounder,the spore morphology rule is closer to the circular,and the mutant strain grew faster than the original strain.
