摘  要:目的:探究迷迭香提取物(RE)的抗氧化能力及其作用机制。方法:以2 d龄Oregon K野生型雄性果蝇为试验模式生物,饲喂添加不同质量浓度(0,0.2,0.5,1.5 mg/m L)迷迭香提取物的培养基,采用生存试验与急性试验研究迷迭香提取物对果蝇寿命的影响;测定果蝇体内超氧化物歧化酶(SOD)与过氧化氢酶(CAT)活力及氧化产物丙二醛(MDA)的含量;用Real time-PCR法测定果蝇体内抗氧化基因SOD、CAT、MTH(Methuselah)m RNA表达量的变化。结果 :给予迷迭香提取物后,果蝇的平均寿命及最高寿命较空白对照组均明显延长,SOD、CAT酶活力及其基因m RNA表达水平显著升高(0.5,1.5 mg/m L,P<0.05),MDA含量极显著下降(P<0.01),同时MTH m RNA表达水平呈下调趋势(P>0.05)。结论 :迷迭香提取物延缓衰老与抗氧化的作用可能通过上调内源性抗氧化酶来实现。Objective: The present study investigated the antioxidant activity of rosemary extract in Drosophila melanogaster and explored its underlying mechanism. Methods: Male flies within 2 d-old were kept on the medium made by rosemary extract with different content of 0, 0.2, 0.5, 1.5 mg/m L. Longevity test and challenge tests were carried out to find the effects of rosemary extract on the lifespan of fruit flies. The antioxidant activities of superoxide dismutase(SOD) and catalase(CAT) and the malondialdehyde(MDA) content in flies were measured. The m RNA expression levels of antioxidant-related genes namely SOD, CAT, Methuselah(MTH) were measured by Real time-PCR(RT-PCR).Results: Rosemary extract could prolong the mean lifespan and maximum lifespan of fruit flies. The enzyme activity of SOD and CAT increased significantly(0.5, 1.5 mg/m L, P〈0.05) and MDA content decreased significantly(P〈0.01), accompanied by up-regulating gene expression of SOD, CAT and down-regulating of MTH(P〉0.05). Conclusion The rosemary extract exerted the antioxidant activities most likely through up-regulation of endogenous antioxidant enzymes.
