摘  要:目的:采用响应面法优化柠条锦鸡儿中总黄酮超声辅助提取工艺,并研究柠条锦鸡儿总黄酮的体外抗氧化活性。方法:在单因素实验的基础上,以超声时间(A)、料液比(B)、乙醇浓度(C)为自变量,柠条锦鸡儿总黄酮得率为因变量,运用Box-Behnken设计-响应面法优化柠条锦鸡儿总黄酮超声辅助提取工艺。同时,测定柠条锦鸡儿总黄酮对四种自由基的清除作用。结果:柠条锦鸡儿总黄酮较优提取条件为:超声时间43 min、料液比1∶30 g/m L、乙醇浓度75%,在此条件下柠条锦鸡儿总黄酮得率可达3.743%,RSD为0.94%。体外抗氧化性实验结果表明,柠条锦鸡儿总黄酮对DPPH·、羟基自由基(·OH)、ABTS+自由基以及超氧自由基(O-2·)均有一定的清除效果,具有较为优良的抗氧化活性。结论:Box-Behnken设计-响应面法优化柠条锦鸡儿总黄酮提取工艺方法简便,可预测性较好;柠条锦鸡儿总黄酮具有体外抗氧化活性,值得进一步研究和利用。Objective: To investigate the ultrasonic-assisted extract on total flavonoids from Caragana korshinskii kom and evaluate its in vitro antioxidant activity.Methods.On the basis of single-factor test,the ultrasonic-assisted extraction process parameters including the ratio of ultralonic time ( A), material- to- liquid ratio ( B ), ethanol concentration(C) of total flavonoids from Caragana korshinskii kom were optimized by the Box-Behnken design and response surface method,and the yield of total flavonoids was used as the responsive values.Meanwhile,four kinds of free radical scavenging effect of total flavonoids from Caragana korshinskfi kom were determined.Results: The optimum conditions for total flavonoids of Caragana korshinskii kom were as follows, ultrasonic time was 43 rain,material-to-liquid ratio was 1:30 g/mL, and ethanol concentration was 75%. Under these conditions, the yield of total flavonoids could reach 3.743%, and RSD was 0.P4%. Result of antioxidation showed that total flavonoids from Caragana korshinskfi kom had a certain scavenging effect on DPPHo, hydroxyl free radical( ·OH), ABTS+ free radical and superoxide free radical(O2- ),so it had good antioxidant activity.Conclusion.The Box- Behnken design and response surface analysis could well optimize the ultrasonic-assisted extraction of total flavonoids from Caragana korshinskfi kom and had good predictability. The total flavonoids from Caragana korshinskii kom had antioxidant activity in vitro, so this plant was worth further research and utilization.
