摘  要:本文从NF-κB信号通路出发,探讨益生菌混合物对人工诱发溃疡性结肠炎小鼠的治疗作用,为治疗人类肠炎提供一种安全可靠的方法。给予溃疡性结肠炎小鼠一定剂量的益生菌混合物,通过Western-blotting、ELISA实验法,分别测定小鼠组织及血清中关键标志物中的表达水平。结果表明:与肠炎空白组小鼠相比,益生菌联合疗法使UC小鼠血清IL-1β、IL-6、TNF-α水平显著降低(p<0.05),并降低NF-κB的蛋白表达,增加IκB的蛋白表达。益生菌混合物通过抑制NF-κB信号通路的活化从而发挥抗溃疡性结肠炎功效,益生菌混合物抑制了NF-κB与IκB的分离,阻塞游离的NF-κB进入细胞核,从而无法调控IL-1β、IL-6、TNF-α等炎性细胞因子的表达,最终抑制机体过度的活化。益生菌混合物对溃疡性结肠炎具有较好的治疗作用,具有应用于临床医学的潜力。Based on the NF-Kappa B signaling pathway, the effect of probiotic mixture on the artificial ulcerative colitis mice was investigated, which provided a safe and reliable way for the treatment of human gastrointestinal Inflammation.Ulcerative colitis mice were provided with some probiotic mixture,and expression of crucial marker were determined by Western-blotting and ELISA tests.Results showed that the serum levels of IL-6, IL-1β and TNF-α were significantly decreased(p 〈0.05) ,and that the protein expression of NF-KB was decreased and the protein expression of IκB was increased, in the probiotics group.Oral administration of L.plantarum, L.acidophilus and L.helveticus suppressed NF-κB signaling pathway and had the anti-colitis effects. In detail, the probiotics inhibited the dissociation of NF- κB from IKB-α, blocking the translocation of NF- κB into the nucleus, down- regulated expression levels of the cytokine gene and suppressed the over-activation of the mice.Thus, probiotic mixture had good effect on the ulcerative colitis and had the potential to be applied into the clinical medicine.
