摘  要:通过单因素和正交试验,研究不同因素对复合蛋白酶水解油菜蜂花粉效果的影响,优化了复合蛋白酶水解的最佳工艺、蜂花粉多肽饮料的最佳配方并分析了蜂花粉多肽饮料的抗氧化活性。酶解的最佳工艺条件为:10.0%油菜蜂花粉浆液,复合蛋白酶添加量3.5%(碱性蛋白酶与中性蛋白酶质量比=2∶1),p H 8.5,酶解温度55℃、时间200 min;在该条件下,多肽的平均得率74.37%。油菜蜂花粉多肽饮料制作的最佳配方为:以油菜蜂花粉蛋白酶解液100%为基准,分别添加0.10%柠檬酸,4.00%蔗糖,0.25%黄原胶。利用该配方制作的产品口感清爽,酸甜适中,味感协调,柔和;浅黄色,质地均匀,无颗粒或者絮状沉淀物;有油菜蜂花粉特有的香气,无异味;多肽含量1.63%(m/V),各项理化指标和微生物指标均符合相应国家标准;该产品具有良好的抗氧化活性,对清除羟自由基和超氧阴离子分别为85.28%、46.52%。In this study, a combination of Alcalase and Neutrase was used to hydrolyze rape pollen to obtain pol- ypeptide and develop rape pollen polypeptide beverage. With the peptide yield as the index, single-factor and orthog- onal experiments were employed to optimize the hydrolysis conditions. Results indicated that the optimal processing conditions for hydrolyzing rape pollen were as follows: 10.0% rape pollen, 3.5% Alcalase and Neutrase (2:1 ) , pH values 8.5, temperature 55℃. Under these conditions, the polypeptide yield reached to 74.37%. The optimum for- mula of rape pollen polypeptide beverage was additonal 0.10% citric acid, 0.10% sugar, 0.25% xanthan gum. The beverage had a nice taste and special flavor of rape pollen and its polypeptide content (m/V) was more than 1.53% , all the physicochemical and health indices met national standards. Moreover, the product showed significant antioxidant activity with, the scavenging rate of hydroxyl radical and superoxide anion radical of 85.28% and 46.52% , respectively.
