摘 要:采用响应面法比较了微波辅助反胶束萃取和超声波辅助反胶束萃取牡丹籽蛋白工艺。得到微波辅助反胶束萃取牡丹籽蛋白的最佳条件为微波时间10.20 min、微波温度47.66℃、微波功率400 W,此条件下牡丹籽蛋白萃取率为48.22%;超声波辅助反胶束萃取牡丹籽蛋白的最佳条件为超声时间60 min、超声温度30℃、料液比1∶50,此条件下牡丹籽蛋白萃取率为84.33%。通过比较牡丹籽蛋白萃取率,得出超声波辅助反胶束萃取牡丹籽蛋白的效果比微波辅助反胶束萃取的效果好。利用反胶束体系萃取牡丹籽蛋白工艺,不仅蛋白质萃取率高、工艺简单,并且避免传统提取过程蛋白质易变性的问题。Microwave - assisted reverse micelle extraction and ultrasound - assisted reverse micelle extrac- tion of peony seed protein were compared by response surface methodology. The optimal microwave - as- sisted reverse micelle extraction conditions of peony seed protein were obtained as follows : microwave time 10.20 min, microwave temperature 47.66 ℃ and microwave power 400 W. Under these conditions, the ex- traction rate of peony seed protein was 48.22%. The optimal ultrasound - assisted reverse micelle extrac- tion conditions of peony seed protein were obtained as follows : ultrasonic time 60 rain, ultrasonic tempera- ture 30 ~C and ratio of material to liquid 1 : 50. Under these conditions, the extraction rate of peony seed protein was 84.33%. By comparing the extraction rates of peony seed protein, the effect of ultrasound - assisted reverse micelle extraction of peony seed protein was better than that of microwave - assisted re- verse micelle extraction.
