摘 要:为了建立适合河豚毒素免疫层析检测的前处理方法,采用超滤技术对前处理方法进行了优化,并对建立的磁性免疫层析试纸条进行了评价。研究结果显示,采用截留分子质量为10 ku的超滤处理,4000 r/min离心10 min的处理是最优的,可以降低溶液中的蛋白浓度,加标回收率在92.4%~126.0%之间,适合免疫层析检测。为河豚毒素的快速现场检测提供了前处理技术参考。In order to establish a suitable pretreatment method for immunochromatographic detection for tetrodotoxin,in this study,the ultrafiltration technology was applied to optimize the pretreatment. The magnetic immunochromatographic test strip established by our laboratory was used for evaluation. The results showed that the optimal treatment of centrifugation at 4000 r / min for 10 min with a cut off molecular of 10 ku. This method was suitable for immunochromatographic assay with good recovery( 92. 4%- 126. 0%). Also,the concentration of protein in the solution reduced after pretreatment. This study provides technical support for rapid on-site test of tetrodotoxin.
