摘  要:华根霉发酵可产酯化酶,在一定条件下能够催化己酸与乙醇反应生成己酸乙酯,以合成的己酸乙酯含量为指标对己酸乙酯的酯化合成条件进行单因素实验与正交实验,确定了各因素的最佳水平。通过优化确立了酯化液的反应体系:以老窖泥富集液与无水乙醇按照10:90的比例配成100 mL反应体系,添加3%的己酸,再以100 g/L华根霉麸曲作为粗酶制剂添加入体系中,置于100 mL的具塞三角瓶中,25℃下,反应25 d,合成的己酸乙酯可达2302 mg/100 mL。Esterifying enzyme produced by the fermentation of Rhizopus chinensis can catalyze hexanoic acid and ethanol to produce ethyl caproate under certain conditions. The optimum synthesis conditions of ethyl caproate were determined by single factor test and orthogonal test. The best reaction system of esterifying liquid were determined as follows: 100 mL reaction system was prepared by mixing aged pit mud enrichment liquid with anhydrous ethanol at the ratio of 10:90, adding 3 % hexanoic acid, and adding 100 g/L Rhizopus chinensis starter as the crude enzyme. The reaction system was placed into 100mL flask with a plug, after 25 d reaction at 25 ℃, the yield of ethyl caproate reached up to 2302 mg / 100 mL.
