摘 要:以福建霞浦海带为研究对象,对超声辅助醇提甘露醇工艺条件进行优化。研究超声时间、超声温度和液固比对甘露醇得率的影响,显著性为:超声温度>液固比>超声时间。并采用Box-Behnken Design中心试验组合设计优化确定最佳提取工艺参数为:以95%乙醇为提取剂,超声功率300 W,液固比52︰1 mL/g,超声时间33 min,超声温度60℃。在该条件下,得率为16.36%。甘露醇的提取受海带溶液黏度影响,采用酸水解前处理降低黏度,释放甘露醇。结果表明:当用2 mol/L盐酸前处理7 h后进行超声醇提,甘露醇得率提高到17.67%。海带酸解预处理与超声辅助醇提甘露醇工艺研究,提高了海带加工的经济效益,为海带高值化利用提供参考。Laminaria japonica produced in Xiapu County of Fujian Province was taken as the research object, and the processing ofmannitol extraction by ultrasound-assisted ethanol was optimized. The effects of ultrasonic time, ultrasonic temperature and solid-liquid ratio on mannitol yield were investigated, and the ranking of significant factors was temperature〉 solid-liquid〉 time. The Box-Behnken Design method was used to optimize extraction parameters. The results revealed that the highest mannitol yield of Laminariajaponica was 16.36%, which obtained with optimum parameters as follows: the exlraction agent was 95% ethanol, the liquid-solid ratio was 52 : 1 mL/g, the time was 33 min, and the temperature was 60 ℃. The extraction of marmitol affected by the solution viscosity, therefore the pretreatment method by acid was adopted to reduce the viscosity for releasing mannitol. Eventually, the yield of mannitol would increase to 17.67%, after pretreated by 2 mol/L HCl solution for 7 h. This study would improve economic efficiency of Laminariajaponica processing, and provide reference for its efficient utilization.
