摘 要:研究高产卡那霉素B产业化工程菌的构建.以主产氨甲酰卡那霉素B工程菌——黑暗链霉菌S.tenebrarius312为出发菌株,进一步敲除氨甲酰磷酸转移酶基因tobZ,获得直接产卡那霉素B的工程菌ST314.对工程菌ST314的生物合成潜力进行研究,在200L发酵罐上进行放大实验,测定各参数,并绘制发酵代谢曲线.结果表明,该菌株遗传特性稳定,产抗能力得到有效恢复,提高幅度接近2倍;该工程菌产卡那霉素B的能力达到4500 U/mL.黑暗链霉菌ST314的研究可以向产业化生产转移.The article studies the construction of high yield Kanamycin B industrialized engineering bacterial.Taking Streptomyces tenebrarius S.tenebrarius 312,which mainly produces carbamoy kanamycin B engineering bacteria,as the original strain,it further removes Carbamyl phosphate transferase genes tobZ and obtains the engineering bacteria ST314 which can directly produce kanamycin B,the article also studies the biosynthesis potential of engineering bacteria ST314,conducts scale-up experiment on 200 Lfermentation tank,measures parameters,and draws fermentation-metabolism curve accordingly.The result shows that,the bacterial strain is stable in hereditary character,the ability of anti-body producing is recovered effectively to around 2times higher,and the ability of producing kanamycin B of the engineering bacteria reaches 4500 U/mL.The making of streptomycete ST314 can be shifted into industrialization.
