摘 要:[目的]本文旨在探究NaCl与CaCl_2处理对西兰花芽苗中硫苷(GLs)和异硫氰酸盐(ITCs)含量以及黑芥子酶(MYR)活性的影响,从而优化出最适的盐浓度富集西兰花芽苗中的GLs和ITCs。[方法]利用HPLC法测定GLs和ITCs含量,并通过测定MYR与底物硫苷反应后生成的葡萄糖含量表示MYR活性。用单因素试验确定NaCl与CaCl_2浓度范围,然后进行响应面分析,优化出NaCl与CaCl_2处理浓度。[结果]NaCl和CaCl_2处理均显著增加西兰花芽苗中GLs和ITCs含量,但对MYR活性无显著影响;随着NaCl浓度的增加,GLs和ITCs含量均呈先增加后减少的趋势,而CaCl_2处理浓度对GLs和ITCs含量均无显著影响。174.09 mmol·L^(-1)的NaCl和10.48 mmol·L^(-1)的CaCl_2为最佳联合施用浓度,在此条件下,西兰花芽苗中GLs和ITCs含量最高,分别达到每10株含5.57μmol和3.54μmol。[结论]用适宜浓度的NaCl与CaCl_2处理西兰花芽苗可使GLs和ITCs得以富集。[Objectives] This work is aimed at studying the influence of NaCl and CaCl2 treatment on glucosinolates (GLs),isothiocyanates (ITCs) contents,and myrosinase(MYR) activity in broccoli sprouts.Then,the optimum salt concentrations for enhancing the GLs and ITCs content of broccoli sprouts were optimized.[Methods] The contents of GLs and ITCs were measured by liquid phase and MYR activity was determined according to the glucose content produced by the reaction between MYR and GLs.Single factor experiments were performed to determine the concentration range of NaCl and CaCl2,and then the response surface was used to optimize the concentration of NaCl and CaCl2.[Results] The treatment of NaCl and CaCl2 significantly increased the content of GLs and ITCs,but had no significant difference on the MYR activity.With the increase of NaCl concentration,GLs and ITCs contents increased continuously and then decreased,while they showed no significant difference under CaCl2 treatment.The 174.09 mmol·L^-1of NaCl combined with 10.48 mmol·L^-1 of CaCl2 treatment was optimum for GLs and ITCs accumulation.Under this condition,both GLs and ITCs contents reached the peak value,which was 5.57 μmol per 10 sprouts and 3.54 μmol per 10 sprouts,respectively.[Conclusions] Suitable concentrations of NaCl and CaCl2 could increase the GLs and ITCs contents of broccoli sprouts.
