摘  要:采用酶法辅助超高压均质技术提取裂殖壶菌油脂,考察了破碎次数、菌体浓度、破碎压力、酶用量对毛油提取率的影响。结果表明:在菌体浓度为200 g/L(湿重),破碎压力为200MPa,破碎1次后,添加2%的液体碱性蛋白酶,在其最适条件下酶解反应3 h后,毛油提取率可达92.12%,该复合法比单一的超高压均质技术提取率高出12.12%,为其工业化应用提供了参考。Lipid extraction from Schizochytrium sp. by enzyme assisted with ultra-high pressure homogenization was studied. The times of cell disruption, cell concentration, pressure of cell disruption and dosage of enzyme was investigated. The results showed that the extraction rate of crude oil was higher when the cell was enzymatically hydrolyzed after cell concentration(wet weight) of 200 g/L disrupted one time by 200 MPa; the highest extraction rate of crude oil(up to 92.12%) was achieved with alkaline protease dosage of 2%, enzymatic hydrolysis for 3 h at the optimal conditions. The extraction rate of composite method increased by 12.12% as compared with the single ultra-high pressure homogenization, provided reference for its industrial application.
