摘 要:选择特定萌发阶段马铃薯制备马铃薯PDA培养基,研究其对霉菌的培养效果。通过测定不同芽长马铃薯还原糖含量和淀粉酶活性,选择还原糖含量最高阶段马铃薯为原料制备马铃薯PDA培养基,分别接种黑曲霉、毛霉和青霉,并比较其培养效果。结果表明,马铃薯芽长2 cm时还原糖含量最高,达到37%;淀粉酶活性也达到最高,为5.2 U/mg。发芽马铃薯PDA培养基可以促进霉菌生长加速,菌落覆盖范围更大;青霉在培养72 h后出现分泌现象。马铃薯发芽过程还原糖含量和淀粉酶活性升高;发芽马铃薯PDA培养基可以加快霉菌生长速度。Selecting the specific germination stage of potato to production of PDA culture medium, and study the effect of the culture of the moulds in above PDA culture medium. The content of reducing sugar and the amylase activity of different shoot length of germination potato are determined. Select the highest stage of reducing sugar content of potatoes as a raw material for preparing potato PDA medium, axe inoculated with Aspergillus niger, Hairy mould and Blue mould, and compare their culture effect with CK. The highest reducing sugar content of potato with shoot length 2 cm is 37%, and the amylase activity is the highest, which is 5.2 U/mg. Germination of potato PDA culture medium can accelerate the growth of mold, the mould colony covering a larger than CK. The secretion phenomenon of Blue mould in germination potato PDA after 72 h culture appeared. The reducing sugar content and amylase activity of germination potato are increased, and the growth rate of mould could be accelerated with the germination potato PDA culture.
