摘 要:建立柱前在线衍生—高效液相色谱法(HPLC)测定发芽糙米中γ-氨基丁酸(GABA)含量的方法。样品以料液比1:5、超声功率(占总功率)60%、在30℃下超声30 min,提取2次。衍生试剂OPA与样品在线混合10次、衍生2 min,用Agilent Eclipse Plus C18柱分离(流速1 mL/min,检测波长338 nm,柱温40℃)。GABA在5~50 mg/L范围内呈良好线性关系,相关系数大于0.999 0;最低检出限0.33 mg/L,最低定量限1 mg/L,方法添加回收率94.80%~98.40%,RSD为1.82%~2.58%。该方法简单快速、灵敏度高、精确,适合发芽糙米中GABA的检测。A HPLC method with online pre - column derivatization for determination of γ- amino butyricacid (GABA) in germinated brown rice was established. The germinated brown rice sample was extracted two times by ratio of solid to liquid 1:5 , ultrasonic power account for 60 % of total power, at 30℃ for 30 min. Online pre - column derivation 2 min were reacted by using orthophthalaldehyde ( OPA ) derived re-agents and sample after mixing 10 times. The sample was separated on an Agilent Eclipse Plus C 18 column ( at 1.0 mL /min, 338 n m and 40℃ ). Good linearity was observed in the range of 5 -50 m g / L with the correlation coefficient 〉 0. 999 0; m i n imum detection limit 0. 33 m g/ L ,minimum quantitative detec-tion limit of 1mg/ L,fortified recovery 94.80% -98.40%,RSD 1.82% -2.58%. The proposed meth-od is simple, rapid, sensitive, accurate and suitable for the detection of GAB A in germinated brown rice.
