摘 要:建立高效液相色谱法快速测定彩色马铃薯黑美人、红宝石中维生素C含量的方法。样品采用2%草酸提取,利用高效液相色谱技术,以0.05 mol/L,pH2.5的磷酸-甲醇(90∶10)缓冲液为流动相,通过C_(18)色谱柱分离,采用紫外检测器,在245 nm波长下测定维生素C的吸光值,将吸光值与标准品进行比较,保留时间可以对其进行定性,以峰面积进行定量。结果表明,方法的线性范围0.02 g/L^0.1 g/L,检出限为0.002 g/L,线性回归方程为yb=5 622 000x+69 538,相关系数为0.999 9。通过回收实验,黑美人与红宝石马铃薯V_C测定回收率分别为97.61%~99.81%,97.28%~99.82%,相对标准偏差分别为0.90%、1.02%。用高效液相色谱法、紫外分光光度法、碘滴定法分别测定黑美人中V_C含量分别为(21.27±0.19)、(19.92±0.13)、(23.31±0.11)mg/100 g,红宝石中V_C含量分别为(18.28±0.19)、(17.68±0.14)、(21.76±0.09)mg/100 g。与甘肃省农业科学院农业测试中心检测结果相比较(黑美人21.73 mg/100 g;红宝石18.74 mg/100 g),表明高效液相色谱法获得结果更准确可靠。本研究所建立的高效液相色谱法可快速、灵敏、准确、稳定的对彩色马铃薯中的维生素C进行测定,同时适用于其它同类果蔬(草莓、刺梨、青菜等)中V_C含量的测定。A rapid method for determination of vitamin C in colorful potato species Solanum tuberosum black beauty and red ruby was established by high performance liquid chromatography (HPLC). Vitamin C in potatoes was extracted by 2 % oxalic acid, then separated through a C18 chromatographic column with the phosphoric acid - methanol buffer (0.05 mol/L, pH2.5 ) as the mobile phase. Thereafter, the vitamin C was detected by ul traviolet-visible detector at the wavelength of 245 nm. The absorbance in samples was compared with the standard substance, which qualitative determination was according to the retention time, and quantitative determination was according to the peak area. The results showed that the linear range of the present method was 0.02 g/L- 0. 1 g/L, the detection limit was 0.002 g/L, and the linear equation was yb =5 622 000x + 69 538, the related coefficient was 0.999 9. After the recovery experiment, the recoveries were 97.61%-99.81%, 97.28 %-99.82 %, respectively, and the relative standard deviation were 0.90 %, 1.02 % ,respectively. The Vc content in black beauty was (21.27±0.19), (19.92±0.13), (23.31±0.11) mg/100 g by HPLC, UV spectrophotometry, Iodomet- ric method, respectively. And (18.28±0.19), (17.68±0.14), (21.76±0.09) mg/100 g in red ruby, respectively. Compared with the results from agricultural test center of Gansu academy of agricultural sciences (21.73 mg/100 g in black beauty; 18.74 mg/100 g in red ruby), the results of HPLC was preciser than the other two methods. The method can well be applied in the determination of vitamin C content with the features of rapid, sensitive, exact and stable, and applicable to detection of vitamin C in fruits and vegetables, such as strawberry, roxburgh rose and green vegetables.
