摘  要:采用超声波辅助法提取鸢尾(Iris tectorum Maxim)叶中异黄酮,运用正交试验优化其提取工艺,并运用自由基清除试验考察鸢尾叶异黄酮的抗氧化活性。研究结果表明,影响异黄酮提取率大小的因素依次为:乙醇体积分数>超声功率>超声时间>料液比,异黄酮提取的最佳工艺条件为:乙醇85%,料液比1︰25(g/mL),超声功率90W,超声时间50 min。此条件下鸢尾叶中异黄酮的得率为15.62 mg/g。此外,鸢尾叶异黄酮对DPPH及羟自由基均显示显著清除活性,表明其具有较强抗氧化活性。Ultrasonic-assisted extraction method was adopted to extract the isoflavones from leaves oflris tectorum Maxim, and an orthogonal design was performed to determine the optimum extract conditions. Also the in vitro antioxidant activity of the I. tectorum isoflavones was evaluated by using free radicals scavenging assays. The results showed that the factors affecting the isoflavones extraction rate descending order were as follows: ethanol concentration, ultrasonic power, ultrasonic time, and material to liquid ratio. The optimum extract conditions were 85% ethanol, material to liquid ratio 1 : 25 (g/mL), ultrasonic power 90 W, and ultrasonic time 50 min. Under these conditions, the yield of total isoflavones was 15.62 mg/g. It was also found that the isoflavones could significantly scavenge DPPH and hydroxyl radicals in a concentration-dependent manner, which indicated that I. tectorum leafisoflavones had strong in vitro antioxidant activity.
