摘 要:目的探究魔芋葡甘聚糖对高尿酸血症大鼠的改善效应;方法用腺嘌呤与氧嗪酸钾联合饲喂酵母饲料的方法造成高尿酸血症的模型,56只雄性大鼠随机分为空白对照组、模型对照组、阳性对照1别嘌呤醇组、阳性对照2苯溴马隆组、高中低KGM组,喂养4w,每周测定一次指标,实验测定血清指标:尿酸(uric acid,UA)、肌酐(creatinine,SCR)、尿素氮(urea nitrogen,BUN)、总超氧化物歧化酶(total superoxide dismutase,T-SOD)活力、过氧化氢酶(catalase,CAT)活力、丙二醛(malondialdehyde,MDA)、总谷胱甘肽(total glutathion,T-GSH)、氧化型谷胱甘肽(oxidized glutathione,GSSG)、脏器指数以及24h尿量和尿酸排泄量,并做了肾脏的病理学观察。结果别嘌呤醇、苯溴马隆与KGM分别不同程度地降低了高尿酸血症模型大鼠的血清尿酸、肌酐和尿素氮水平;与正常组相比,所有试验物均能减少高尿酸血症肾病大鼠的排尿量;KGM中剂量组的MDA含量最接近正常组;KGM中剂量组的血清SOD值显著(P<0.05=)低于模型组;各组大鼠的总谷胱甘肽含量无显著(P>0.05)变化;与正常组相比,模型组的GSH/GSSH比值显著(P<0.05=降低;各组大鼠肾脏体积明显大于正常大鼠肾脏;各组大鼠的脾体指数和肺体指数无显著(P>0.05)变化,但各组高尿酸血症大鼠的心体指数、肝体指数和肾体指数显著(P<0.05)上升;KGM高剂量组可有效清除大鼠肾脏中过量产生的尿酸盐结晶。结论本试验高尿酸血症大鼠造模成功,魔芋葡甘聚糖对高尿酸血症大鼠有改善效果。Objective To investigate the effect of konjac glucomannan(KGM) on hyperuricemia in rats. MethodsThe hyperuricemic rat model was established by feeding a yeast combined with adenine and oteracil potassium diet. Fifty-six SD male rats were randomly divided into 7 groups(n=8/group) : blank control group, model group, positive control 1 allopurinol group, positive control 2 benzbromarone group, KGM high-, middle-and low-dose groups, and all diets were fed for four weeks. Serum uric acid(UA), creatinine(SCR), urea nitrogen(BUN), total superoxide dismutase(T-SOD), catalase(CAT), malondialdehyde(MDA), total glutathion(T-GSH), oxidized glutathione(GSSG), the organ index, and 24-hour output of urine and uric acid were measured. Kidney pathological observation was conducted. Results Allopurinol, benzbromarone and KGM reduced the levels of creatinine and urea nitrogen in hyperuricemic rats. Compared with the normal group, the urine output in rats with high uric acid nephropathy were reduced in all the intervention groups. MDA content in medium KGM group was the closest to the normal group. The serum SOD activity in medium KGM group was significantly(P〈0.05) lower than in the model group. The content of glutathione did not changed significantly(P〉0.05). Compared with the normal group, GSH to GSSH ratio in the model group were significatnly(P〈0.05) decreased. Kidney volume of the rats in hyperuricemic groups was much higher than that of the normal rats. There was no remarkable(P〉0.05) difference in spleen and lung indexes. However, the heart, liver and kidney indexes in hyperuricemic rats increased significantly(P〈0.05). High-level KGM was effective in removing lithate crystals overproduced in the kidney. Conclusion The model of hyperuricemia rats was successfully established, The KGM has a positive effect on the hyperuricemia in rats.
