摘 要:目的:为探讨壳聚糖对病原菌的抑制作用及对拮抗菌的影响,以期为拮抗菌Bacillus amyloliquefaciens BA-16-8应用于苹果采后的防腐保鲜提供理论基础。方法:以日本红富士为实验材料,采用体外及体内实验方法,研究不同浓度的壳聚糖对扩展青霉(Penicillium expansum)的抑制作用及解淀粉芽孢杆菌BA-16-8对扩展青霉产毒的影响。结果:浓度2%的壳聚糖溶液抗P.expansum和青霉病的能力最强,且不会影响拮抗菌B.amyloliquefaciens BA-16-8的生长。拮抗菌B.amyloliquefaciens BA-16-8的菌悬液及无细胞发酵液均可抑制P.expansum,且菌悬液的处理效果要优于无细胞发酵液。抑制产毒实验结果表明,B.amyloliquefaciens BA-16-8菌悬液及其无细胞发酵液均可显著抑制P.expansum发酵液中展青霉素的积累,但对单纯的展青霉素并无降解作用。结论:B.amyloliquefaciens BA-16-8同壳聚糖联合作用可抑制扩展青霉的活性及产毒能力。Objective: In order to discuss the inhibitory effect of chitosan on both pathogenic bacteria and biocontrol angent, and to provide theoretical basis for application of Bacillus amyloliquefaciens BA-16- 8 in the antisepsis and fresh-keeping for postharvest apple. Method: Taking Fuji apple as experimental meterial, the inhibition effect on Penicillum expansum under different concentration of chitosan and patulin (PAT) removal ability by different concentration of fermentation broth of B.amyloliquefaciens BA- 16-8 was determined in vivo and vitro. Result: As was shown in the result, the ideal concentration of chitosan was 2%, which could not only exbihit strong anti-fungal ability against P. expansum but also don't inhibit growth of B. amyloliquefaciens BA-16-8. Moreover, both the fermentation broth and cell- free fermentation broth of B. amyloliquefaciens BA-16-8 could inhibit the growth of P.expansum, whereas the fermentation broth showed the stronger anti-fungal ability. Antifungal and patulin (PAT) removal experiments revealed that the cell-containing and cell-free fermentation broths of B.amyloliquefaciens BA- 16-8 could significantly inhibit the PAT accumulation in fermentation broth of P. expansum. However, B. amyloliquefaciens BA-16-8 couldn't degrade the patulin. Conclusion: Both the mycelia ability and patulin- producing ability of P. expansum would be significantly inhibited by combining the concentration of 2% (w/v) chitosan with fermentation broth of B. amyloliquefaciens BA-16-8.
