摘 要:食源性致病菌的快速检测一直是保障食品安全的关键。沙门氏菌(Salmonella)是重要的食源性致病菌之一。本研究以沙门氏菌人工污染的3种样品为对象,对一种基于环介导等温荧光扩增技术的沙门氏菌快速诊断试剂盒进行了灵敏度和准确性评估;再以该试剂盒对市售食品样品和临床腹泻样品适用性进行验证。结果表明,该等温荧光定量沙门氏菌快速检测试剂盒的检测限为10~3 CFU/mL,准确性96.7%以上。所有不同样品的结果显示:阴性样本的假阳性率分别为0.0%、3.3%和3.3%;阳性样本检出率分别为97.9%、100.0%和65.7%(含低浓度细菌污染样品)。该试剂盒具有检测周期短、操作简便、适用性广等特点。Rapid detection of foodborne bacteria is a key for food safety. Salmonella is one of the important foodborne pathogenic bacteria. In this study, the artificially contaminated samples with Salmonella were used to evaluate the sensitivity and accuracy of a retail kit based on isothermal amplification. Three kinds of samples were detected, including bacteria culture, food and clinical samples. The results demonstrated that the detection limit of the kit was 10a CFU/ml and the accuracy rate was over 96. 7%. In addition, the false-positive rate in all negative samples was 0. 0%,3.3% and 3.3% respectively. The positive rate in all positive samples was 97.9%, 100. 0% and 65.7% (including low detection limit samples)respectively. In conclusion, the advantage of the kit was less time consuming, simple protocol and wide adaptability.
