摘  要:为获得繁殖快且药用价值高的铁皮石斛,以多糖含量为筛选指标,选择原球茎为载体,运用Plackett-Burman设计从7种影响原球茎多糖含量的培养基成分中确定硝态氮/铵态氮分子比、蔗糖浓度和苄氨基腺嘌呤(BA)浓度为其中最重要的影响因素。进一步设计最陡爬坡实验逼近中心点并进行Box-Behnken Design响应面方法分析,得出回归模型存在的最优结果为:当硝态氮/铵态氮分子比、蔗糖浓度、BA浓度分别为1.90∶1,29.1 g/L,1.65 mg/L时,多糖含量由优化前的25.36%提高到了33.95%,优化效果显著。To obtain fast-breeding and high medicinal-value.D, officinale, polysaccharide content was identifying indicator. Taking PLB as a carrier,Nitrate/ammonium molecule ratio, sucrose and BA concentration were recognized the most important factors in seven medium compositions affecting PLBs polysaccharide content through the Plackett-Burman design.The steepest ascent experiment and further Box- Behnken design response surface methods were designed, optimal results of the presence of regression model were as follows:nitrate/ammonium molecule ratio, sucrose and BA concentration were 1.90:1,29.1 g/L, 1.65 mg/L. Under these conditions, the mass fraction of the polysaccharide increased 33.95% with a significant improvement compared with the value of 25.36% before optimization.The optimization effect was remarkable.
