摘 要:以水解度为指标,通过单因素实验和正交实验,确定了深海鳕鱼皮胶原蛋白粉的酶解最优工艺参数,同时对最优参数酶解所得产物开展氨基酸组成分析。结果表明:中性蛋白酶、胰蛋白酶、复合蛋白酶、风味蛋白酶4种酶制剂中,胰蛋白酶酶解深海鳕鱼皮胶原蛋白粉溶液的效果最佳,最佳酶解工艺条件为固液比1:3、初始pH8、酶解温度48℃、酶用量0.8%、酶解时间4h。用氨基酸分析仪对酶解产物的氨基酸进行分析发现,酶解产物氨基酸总含量为82.993%,主要含有15种氨基酸,其中包括必需氨基酸6种,分别是苏氨酸、缬氨酸、异亮氨酸、亮氨酸、苯丙氨酸、赖氨酸。15种氨基酸中,苯丙氨酸、亮氨酸、甘氨酸和精氨酸的含量较高。The optimum enzymatic parameters of the hydrolyzed collagen powder were determined by single factor test and orthogonal test. At the same time, the amino acid composition of the obtained product was analyzed. The results showed that, the best experimental results were trypsin in the four experiments of enzymatic hydrolysis of deep-sea cod skin collagen solution, the four kinds of enzyme preparations were prepared by neutral protease, trypsin, complex protease and flavor protease. The optimum parameters were: solid-liquid ratio 1:3, enzyme solution pH 8.0, enzymatic hydrolysis temperature 48 ~C, enzyme 0.8% and time of enzymatic hydrolysis 4 hours. Analysis of the amino acids of the hydrolyzate by amino acid analyzer showed that the hydrolyzate contained 15 kinds of amino acids, including six kinds of amino acids, namely, threonine, valine, isoleucine, leucine, amino acid, lysine. Among the 15 amino acids, the content of phenylalanine, leucine, glycine and arginine were higher.
