摘 要:以鼠尾藻为原料,运用超声辅助提取法对鼠尾藻中岩藻黄素的提取工艺进行研究,通过单因素实验法和响应曲面法确定了工艺参数。通过高效液相色谱法(HPLC)和电喷雾质谱法(ESI/MS)对经过硅胶层析柱分离纯化后的岩藻黄素样品进行分析。实验结果表明,超声提取的最佳工艺是:乙醇体积分数为80%(v/v),超声时间32 min,超声起始温度52℃,液料比10 m L/g,L-抗坏血酸的添加量0.8%,在此条件下提取2次,岩藻黄素粗品得率为(0.455±0.046)mg/g。经硅胶层析柱纯化后的岩藻黄素纯度约86.88%±1.34%,得率为(0.048±0.002)mg/g。采用该法制备高纯度的岩藻黄素工艺简单,产物得率高,为其今后的综合利用提供了实验基础和技术支持。Ultrasonic technology was used to assist the extraction of fucoxanthin from Sargassum thunbergii. The optimum conditions were determined by using single factor experiment and response surface methodology. The high performance liquid chromatography( HPLC) and electrospray ionization mass spectrometry( ESI/MS) method were developed for the analysis of fucoxanthin that was isolated and purified by silica gel column. The experimental results showed that the optimum ultrasonic extraction model was as follows: Ethanol concentration 80%( v/v),ultrasonic starting temperature 52 ℃,extraction time32 min,and solvent to material ratio 10 m L/g,the dosage of L-ascorbic acid 0.8%,extract two times,the fucoxanthin extraction yield was( 0.455 ± 0.046) mg/g.After purification,the purity of the fucoxanthin isolated was about86.88% ± 1.34%,and the extraction yield was( 0.048 ± 0.002) mg/g. This method that prepared the high purity fucoxanthin was simple and the extraction ratio was high,which would provide experimental basis and technical support for its comprehensive utilization in the future.
