摘  要:研究了对铅离子有吸附能力的嗜铅肠球菌JT_1作为体内排铅试剂的排铅机理。建立大鼠铅中毒模型,模型组血铅水平达到600μg/L即造模成功。通过灌胃的方式给予大鼠嗜铅肠球菌JT_1的发酵液,4周后处死,无菌采集结肠进行菌落计数,并进行扫描电子显微镜观察及能谱分析。同时,采集大鼠血液、尿液及粪便测定其铅含量的变化。结肠菌落计数结果显示:铅中毒模型组大鼠结肠乳酸菌总数较其它组少,差异显著(P<0.05);灌喂嗜铅肠球菌JT_1的模型组和对照组菌落总数高于未灌胃空白组和染铅组,差异显著(P<0.05)。结肠扫描电镜研究结果表明:仅能在灌喂嗜铅肠球菌JT_1的模型组和对照组大鼠结肠内发现类似嗜铅肠球菌JT_1的菌体,能谱分析显示模型组大鼠结肠内菌体表面铅含量明显增多,说明铅离子在菌体表面形成金属沉积物。灌胃嗜铅肠球菌JT_1的模型组大鼠的血铅水平均低于灌胃前,差异显著(P<0.05),且该组的粪铅水平显著上升,差异显著(P<0.05)。上述结果表明,嗜铅肠球菌可在大鼠结肠存活,嗜铅肠球菌JT_1与铅离子主要以表面吸附的形式排铅,在灌胃嗜铅肠球菌JT_14周后能有效降低大鼠血铅水平,其排铅的主要途径是随粪便排出。In the study, the lead-resistant bacteria JT1 was used as lead excretion agent to study on its lead-discharging mechanism in rats. Firstly, the lead poisoning model was researched. Then the vivo tests were conducted after blood lead levels above 600 μg/L. Wistar rats were provided with the broth of lead-resistant bacteria JT1 by gavage. And the rats were killed for culturing lactic acid bacteria, observation by scanning electron micrographs and element content of energy spectrum analysis from the colon after four weeks. The blood, urine and feces lead content of rats were measured with Inductively Coupled Plasma Optical Emission Spectrometer. The results of lactic acid bacteria colony counts were that bacteria colony counts of model groups were less than blank groups, and the difference was significant(P〈0.05).The bacteria colony counts of and model group two and blank group two were higher than other groups, and the difference was significant(P〈0.05). Scanning electron micrographs showed the strains of bacteria in the presence of rat colon were similar to JT1, and Element content of energy spectrum analysis showed the adsorption of lead by JT1. It indicated that the lead ions on the cell surface were formed deposit. There were significant differences both in blood lead and fe ces lead of rats after providing with the broth of lead-resistant bacteria JT1 by gavage(P 〈0.05). The results above showed that JT1 can survive in colon of rats, and the bacteria can combine lead by surface adsorption of lead ions in rats’ intestine. The lead-resistant bacteria JT1 can effectively reduce the blood lead levels, and lead was almost ruled out with feces.
