摘 要:以太平洋牡蛎为试验对象,研究不同染Cd浓度和时间条件下太平洋牡蛎体内的Cd含量和P-糖蛋白(P-gp)活性的变化,以及添加P-糖蛋白专一性抑制剂异搏定(Ver)和诱导剂利福平(Rif)对Cd含量的影响。采用火焰原子吸收分光光度法测定Cd含量,排出法测定P-gp的活性变化,结果表明,不同染Cd浓度和染Cd时间对太平洋牡蛎体内的Cd含量及P-gp的活性变化有显著影响,且两者之间的变化相对应,说明P-gp在太平洋牡蛎体内的自身抗Cd机制中发挥作用。添加Ver后,太平洋牡蛎体内Cd含量显著增多,而添加Rif后太平洋牡蛎体内Cd含量下降,说明P-gp在太平洋牡蛎的自身防御机制中发挥重要作用,从而防止机体对外来有害物质的侵入。P-gp抑制Cd在体内的富集,对太平洋牡蛎净化脱Cd,提高其食用安全性具有重要意义。Crassostrea gigas was used as experimental subject, the change of Cd content, P-glycoprotein(P-gp)activity in Crassostrea gigas and adding Verapamil and Rifampicin at different concentrations of Cd and exposure time was studied. The content of Cd and activity of P-gp was determined by the method of flaming atomic absorption and exhaust, respectively. The results showed that Cd concentration and exposure time have great effects on both Cd accumulation and P-gp activity. Verapamil can significantly increase Cd content in Crassostrea gigas, while exposure to Rifampicin has reduced Cd content. It indicated that P-gp played important roles in Cd accumulation, and Rifampicin can effectively reduce Cd content in Crassostrea gigas, which has significant meaning to improve the safety of aquatic products.
