摘  要:【目的】蛇莓为中国民间常用草药,具有良好的生物和药用活性,目前对其抗氧化、降血糖、防治老年痴呆方面研究较少。【方法】以总酚得率为指标,采用响应面法优化蛇莓多酚的微波辅助提取条件;以DPPH自由基清除能力、ABTS+自由基清除能力、铁还原能力为指标,考察其体外抗氧化能力;采用改良的荧光法考察其抑制α-葡萄糖苷酶能力;采用吸收光谱法考察其抑制乙酰胆碱酯酶能力。【结果】微波辅助提取蛇莓多酚的最佳条件为甲醇体积分数67%,料液质量体积比1∶18(W/V),微波功率280 w,微波时间为112 s,在此条件下蛇莓总酚得率为5.21%;蛇莓多酚清除DPPH自由基的IC50值为8.63μg/m L,清除ABTS+自由基的IC50值为32.11μg/m L,铁还原能力为2 190.33μg Fe SO4/mg;抑制α-葡萄糖苷酶的IC50值为223.89μg/m L,低于标准对照物阿卡波糖(310.81μg/m L);抑制乙酰胆碱酯酶的IC50值为202.95μg/m L。【结论】蛇莓多酚有较强的体外抗氧化活性、抑制α-葡萄糖苷酶和乙酰胆碱酯酶的能力,具有开发为天然抗氧化剂和降血糖、防治老年痴呆药物的可能。[ Objective] Duchesnea lndica(DI) is one of the common used herbs in China, it has excellent biological activity and phammcolog- ically active. However, there is less research about anti-oxidant activity, hypoglycemic activity and staving off alzheimer' s of DI. [ Method] Response surface methodology(RSM) was employed to optimize extraction conditions by micro-assisted extraction, with the total phenols yield of DI as response values. The in vitro antioxidant activity of DI polyphenol was evaluated by DPPH free radical scavenging capacity, ABTS + free radical scavenging capacity and ferric reducing ability. The α-glucosidase inhibitory activity of DI polyphenol was evaluated by modified fluorescence method, and the acetylcholinesterase (ACHE) inhibitory activity of DI polyphenol was evaluated by absorption spectros- copy. [ Result] The optimized conditions by micro-assisted extraction were determined as follows: methyl alcohol concentration 67 % , sol- vent-to-solid ratio 1:18 (W/V) , microwave power 280 w, microwave time 112 s. Under these conditions, the total phenols yield was 5.21 %. The IC50 value of DPPH free radical scavenging capacity of DI polyphenol was 8.63 Ixg/mL, and the IC50 value of ABTS + free radical scavenging capacity was 32.11 μg/mL, and the ferric reducing ability was 2 190.33 μg FeSO4/mg. The ICs0value of α-glucosidase inhibi- tory activity was 223.89 μg/mL, renmrkably lower than standard control acarbose( IC50 - 310.81 μg/mL), the ICs0value of AChE inhibito- ry activity was 202.95 μg/mL. [ Conclusion ] DI polyphenol has excellent in vitro antioxidant activity, α-glucosidase inhibitory activity and AChE inhibitory activity. It has the potential to explore as nature antioxidants, hypoglycemic drags and Alzheimer' s drags.
