摘  要:[目的]阐明铜绿假单胞菌prtR严紧调控的分子基础及对绿脓杆菌素表达的影响。[方法]通过5'RACE确定prtR和ptrB的转录起始位点;通过RT-PCR的方法研究prtR的相对表达水平和对绿脓杆菌素表达的影响。[结果]prtR和ptrB的转录起始位点分别位于已预测的结构基因上游-1 bp和-322 bp;另外,PrtR除了通过抑制prtN来抑制绿脓杆菌素的表达之外,它的本底水平的表达对绿脓杆菌素还有一个间接或直接的抑制作用。[结论]造成prtR极低表达水平的原因,除自我抑制外,还存在ptrB对其的转录竞争抑制。此外,prtR独立对绿脓杆菌素的表达有抑制作用。[Objective]The aim of this study is to clarify the molecular basis of tight regulation of prtR,and its effect on the expression of pyocins in Pseudomonas aeruginosa. [Methods]Transcription start sites of prtR and ptrB were determinated by 5'RACE; Relative expression level of prtR and its effect on the expression of pyocins were investigated by Real Time PCR. [Results] Transcription start site of prtR and ptrB are located at 1 bp and 322 bp upstream of structual gene as predicted by published whole genome sequence,respectively. In addition,the basal level expression of PrtR has an inhibitory effect( directly or indirectly) on the expression of poycin genes independent of prt N. [Conclusion] One reason for the extremely low expression level of prtR is due to the competitive inhibition of transcription by ptr B,in addition to self-inhibition. Moreover,prtR can inhibit the expression of Pseudomonas aeruginosa independently.
