鳕鱼皮胶原蛋白多肽的制备及其对小鼠B16黑素瘤细胞的作用

摘　　要：优化鳕鱼皮胶原蛋白的酶解条件以及制备活性多肽酶解液;经超滤得到不同分子量范围的鳕鱼皮胶原蛋白多肽;研究各组分对小鼠B16黑素瘤细胞的生长及黑色素生成的影响。结果表明,鳕鱼皮胶原蛋白多肽制备的最佳酶解条件为:碱性蛋白酶、酶解时间3 h、酶解温度45℃、酶与底物比1∶50(w/w)、pH=9.0;不同分子量范围的鳕鱼皮胶原蛋白多肽对小鼠B16黑素瘤细胞均无毒性作用,并能抑制其黑色素的生成。其中,分子量小于2 000的组分抑制效果最好。In this study,the enzymatic conditions for cod skin collagen were optimized.Four fractions with different molecular weights were obtained from the ultrafiltration of the hydrolysates.The effects of the four fractions on B16 melanoma cell growth and melanogenic activity were investigated.The best hydrolysis technology parameters are obtained as follows: the enzyme is properase E,the hydrolysis time is 3 h,and the enzyme to substrate ratio is 1∶50(w/w),the pH value is 9.0.The four fractions showed no cytotoxicity on B16 melanoma cells and showed some potent inhibition effects on melanin synthesis.The fraction with molecular weight lower than 2 000Da showed the best effect.
