产低温蛋白酶海洋菌筛选、鉴定及性质的初步研究

摘　　要：通过脱脂牛奶平板法从深海海泥中筛选到35株在低温条件(4℃)下具有蛋白酶活性的菌株,并对这35株菌进行了分子生物学鉴定和酶活测定,发现这35株菌分别属于动性球菌属(Planococcus)、芽孢杆菌属(Bacillus)、Fictibacillus、发光杆菌属(Photobacterium)、冷蛇菌属(Psychroserpens)、微小杆菌属(Exiguobacterium)、交替假单胞菌属(Pseudoalteromonas).选择各个属活性最高的菌株为研究对象,以酪蛋白为反应底物对其所产蛋白酶粗酶液酶学性质进行初步研究.结果表明,动性球菌属菌株PL4-1所产蛋白酶酶活最适反应温度为40℃,10℃时可保持50%以上的酶活,在60℃时酶活下降至40%以下,为典型的低温蛋白酶.Thirty five bacteria strains which producing extracellular protease were screened from the deep sea mud by skimmed milk powder plate method and their egzyme activity assays. According to their 16S rDNA sequence analysis of the strains,the results showed that they belonged to Planococcus,Bacillus,Fictibacillus,Photobacterium,Psychroserpens,Exiguobacterium,Pseudoalteromonas respectively. By using the casein as the substrate,the characteristics of the crude protease produced by PL4-1 showed that the optimal temperature was 40 ℃. The relative activity reached more than 50% of the highest activity at 10 ℃ and reduced to 40% below at 60 ℃. These enzymatic properties indicate that the protease belongs to the class of cold-adapted proteases.
