摘  要:研究超声提取总核苷的最佳工艺条件,以核苷功能菌固态发酵物为原料,总核苷提取量为指标,在单因素试验的基础上,选取提取时间、料液比、提取温度、超声频率为自变量,总核苷提取量为响应值,采用Box-Behnken响应面法优化总核苷超声波提取工艺。结果表明,超声波提取总核苷的最优工艺为提取时间54 min,料液比(m L/g)为90∶1,提取温度56~℃,超声频率50 Hz,该条件下总核苷提取量为3.79 mg/g,与预测值相近,说明模型拟合良好,该优化工艺准确可行。The optimum technical conditions of ultrasound-assisted extraction of total nucleosides from solid-state fermentation material of nucleoside functional strain were explored.Total nucleosides extraction amount used as the evaluating index,on the basis of single-factor test,Box-behnken response surface method was used to optimize the extraction with extraction time,solvent/material ratio,extraction temperature and ultrasound frequency as the independent variables,and total nucleosides extraction as the response value.The optimum technical conditions were summed up as follows:extraction time was54min,solvent/material ratio was90∶1,extraction temperature was at56℃,and ultrasound frequency was at50Hz.Under these conditions,total nucleosides extraction was3.79mg/g,close to the predicted value,suggesting that the model fitting was good,and such optimization was accurate and practical.
