摘 要:对气相色谱法测定己酸菌发酵液中己酸等有机酸含量的预处理方法进行了研究,优化了色谱条件,建立了一种快速测定己酸菌发酵液中己酸、丁酸、乙酸等有机酸的方法。己酸菌发酵液经2M硫酸酸化至p H2后经高速冷冻离心,再经微孔滤膜过滤,利用Pure-WAX毛细管色谱柱、以2-乙基丁酸为内标、直接进样气相色谱法定量测定。色谱分析条件为:检测器温度260℃,汽化温度250~℃,柱温150~℃,分流比50∶1,采样时间10 min。研究结果表明,应用该法测定己酸菌发酵液中的己酸、丁酸、乙酸等有机酸含量,RSD为2.22%~2.54%,加标回收率在96.83%~103.71%之间。该方法具有快速方便、定量准确等特点,适合己酸菌发酵液中己酸、丁酸、乙酸等有机酸的检测。The pretreatment for measuring the content of caproic acid and other organic acids in fermented liquid of caproic acid bacteria was studied.Firstly,the fermented liquid underwent2M sulfuric acid acidification till pH=2,then through high-speed freeze centrifugation and micropore filtration,direct sampling GC using Pure-WAX capillary-column and2-ethyl butyric acid as internal standard was performed.The chromatographic conditions were as follows:the detector temperature was at260℃,the vaporization temperature was at250℃,the column temperature was at150℃,the split ratio was50∶1and the sampling time was10min.The results showed that the content of caproic acid,butyric acid,acetic acid and other organic acids could be measured by such method,RSD was2.22%to2.54%,and standard addition recovery was96.83%to103.71%.This method had the advantages such as convenient,fast and accurate determination.
