摘 要:嗜酸乳杆菌-地衣芽孢杆菌融合子分别进行固体培养和液体培养,观察融合子在不同培养基和不同培养条件下的生长状况、个体形态以及菌落形成,探索培养基的种类、pH值、温度、培养方式等对融合子生长量的影响,同时对其产酸性能进行了分析。结果表明:融合子在不同培养基的生长情况为:CM>BPM≈LB>MRS,CM、MRS和LB培养基的适宜初始pH值分别为6、7和5,BPM培养基初始pH值对融合子的生长繁殖影响不显著(P>0.05)。在CM培养基中,37℃更适于融合子生长,其他培养基的最适培养温度均为35℃。除MRS培养基适合隔氧静置培养外,其他3种培养基则配以通氧摇床培养方式。Lactobacillus acidophilus-Bacillus licheniformis fusant was cultured by solid medium and liquid medium respectively.The growth conditions,individual form and colony formation of fusant were observed under the different culture medium and different conditions of culture,the effect of the type of medium,pH value,temperature,training mode were explored,and the acid-production ability was analysed.The results indicated that the growth of fusant in different culture media was as follow in descent:CM>BPM抑LB>MRS.The suitable initial pH of CM,MRS and LB culture media was 6,7 and 5 respectively.The effect of initial pH in BPM on the growth and reproduction of fusant was not significant(P跃0.05).The optimum culture temperature was 37益in CM,and the other three medium(CM,MRS and LB)were 35益.The fusant in the medium of CM,BPMand LB were suitable to shaking culturing with oxygen,but it was suitable to anaerobic culturing in MRS culture.
