南方刺参性腺多肽酶解工艺优化及抗氧化活性分析

摘  要:研究南方养殖刺参性腺多肽的酶解提取条件,并分析南北刺参性腺多肽抗氧化活性。以南移养殖刺参性腺为原料,以酶解多肽得率为衡量指标,选取酶解温度、底物浓度、pH、酶解时间、酶用量等为主要影响因素,在单因素优化基础上应用响应面分析法(RSM)进一步优化,确定刺参性腺多肽酶解工艺。试验结果表明,刺参性腺多肽酶解最佳条件:温度为56℃:,底物浓度11.5%,酶添加量0.5mKat.g-1,pH 7.1,酶解时间4.5 h,此时刺参性腺多肽得率可达到67.97%,与预测值相符。抗氧化试验表明,刺参性腺多肽具有较强的抗氧化能力,南北养殖刺参性腺酶解多肽的抗氧化能力无显著差异。The polypeptides extraction conditions and antioxidant activity of polypeptide in sea cucumber(Apostichopus japonicus)gonads from South China were studied.Protamex was used to hydrolyze the sea cucumber gonads.The peptide yield index was used as the response,and based on the single factor experiments,enzymolysis temperatoe,the substrate concentration,pH,enzymolysis time,and enzyme addition were screened and optimized using response surface methodology(RSM)to determine the optimal conditions of polypeptides extraction.The results showed that the optimal conditions are as follows:temperature at 56℃,11.5%(m:V)of substrate concentration,0.5 mKat/g of enzyme dosage,pH of 7.1,enzymolysis time is 4.5 h.The peptide yield was 67.97%under the optimal conditions,which correspond with the prediction model.The results of antioxidant activity of gonads polypetide showed that sea cucumber gonads polypeptide exhibited effective antioxidant activities,and there is no marked qualitative difference between the North and South cultures.
