摘 要:为探究葡萄藻的保存方法及其与细菌复配后的净水效果,采用固定化、低温、冷冻、液氮超低温等方法保存葡萄藻2 m,以复苏后藻体生物量、比增长率及叶绿素含量为指标筛选葡萄藻适宜的保存方法.通过wadley混剂复配原则筛选葡萄藻与乳酸菌、硝化细菌的复配增效比例.结果表明,葡萄藻中有机物保存的最佳方法为玻璃化超低温法,中短期藻种保存最佳方法为固定化法.单一悬浮藻及藻菌复配体系对人工污水中的氮与磷均有显著清除效果,且去氮的效果优于去磷.硝化细菌对葡萄藻有显著的增效作用,当藻与硝化细菌比为1时,增效效果最佳,其对磷、氨氮、硝酸氮、亚硝酸氮的增效系数SR分别为1.99、1.16、2.35、2.46.乳酸菌对葡萄藻清除磷的效果有一定影响,复配后的清除率低于单一藻体;但对葡萄藻清除氮有一定的增效作用,当藻与乳酸菌比为2时,复配体系对NH_4^+-N、NO_3^--N及NO_2^--N清除率分别达到99.0%、98.2%与35.8%.该研究可为藻菌复合生物微球用于生态净水提供参考配比,同时解决微藻保存过程中易受污染的问题.The aim of the paper was to explore the preservation methods of Botryococcus and its compound with bacteria on purification efficiency of the artificial sewage.Botryococcus was preserved for 2 months by immobilization,low temperature,freezing and liquid nitrogen ultra low temperature,the appropriate preservation methods were selected with the biomass,specific growth rate and chlorophyll content of Botryococcus as indexes after the recovery.The synergistic ratio of Botryococ-cus with lactic acid bacteria and nitrifying bacteria was screened by Wadley mixture principle.The results showed that the best method for preservation of organic compounds in Botryococcus was vitrification,and the best method of preservation in medium and short term was immobilization method.Both the single suspension microalgae and microalgae bacteria com-pound system had significant scavenging effect on nitrogen and phosphorus in artificial waste water,and the effect of nitro-gen removal was better than that of phosphorus removal.Nitrifying bacteria had significant synergic effect on Botryococcus.The synergistic effect was the best when microalgae:bacteria=1:1,and the efficiency coefficients of phosphorus,ammonia nitrogen,nitrate nitrogen,nitrite nitrogen were 1.99,1.16,2.35 and 2.46,respectively.There was a certain effect of lactic acid bacteria on phosphorus removal effect to Botryococcus,and the clearance rate was lower than that of single mi-croalgae.Lactic acid bacteria had a certain synergistic effect to Botryococcus on the removal of nitrogen.When microalgae:bacteria二2:1,the removal rates of NH4+-N、N03_-N and N02_-N were 99%,98.2%and 35.8%,respectively.The study can provide reference rate of Botryococcus and its compound with bacteria on sewage purification,and solve the prob-lem of Botryococcus pollution on the process of saving.
