摘　　要：通过对菌株Bacillus subtilis YKM05发酵产酶条件的优化研究,使其产角蛋白酶活力显著提升.最适产酶诱导物为羽毛粉,最适碳源为玉米粉,最适氮源为豆粕粉,Ca^(2+)、Fe^(3+)对产酶有促进效果,Mn^(2+)、Zn^(2+)、Cu^(2+)对产酶有抑制效果.最佳的培养基组成为:羽毛粉2.0%、玉米粉2.0%、豆粕粉2.0%、磷酸二氢钠0.3%、磷酸氢二钠0.5%、氯化钙0.6%.菌株优化产酶条件为:发酵温度36℃、培养基初始pH值为9、种子接种量为3%、1000 m L三角瓶最佳装液量为300 mL、发酵时间72 h.In this study,the enzyme production fermentation conditions of Bacillus subtilis YKM05 were optimized,and the activity of keratinase was increased significantly.The most suitable enzyme inducers were feather meal,the most suitable carbon source was corn meal,and the most suitable nitrogen source was soybean meal.Ca2+and Fe3+promoted the enzyme production,while Mn2+,Zn2+and Cu2+inhibited the enzyme production.The best medium was 2.0%feather meal,2.0%corn meal,2.0%soybean meal meal,0.3%dihydrogen phosphate,0.5%disodium phosphate,and 0.6%calcium chloride.The optimum enzyme production conditions of the strain as follows:the fermentation temperature is 36℃,the initial pH of the culture medium is 9,the seed inoculation is 3%,the best filling amount of the 1000 mL triangle bottle is 300 mL,and the best fermentation time is 72 h.
