摘 要:建立3,5-二硝基水杨酸(DNS)测定黄水中总糖含量的方法,对检测条件和黄水总糖水解条件进行优化,并进行方法学考查。结果表明,最佳检测条件:显色剂添加量5 mL,显色时间7 min,显色温度90℃;黄水总糖水解条件为:水解时间30 min,盐酸浓度12 mol/L,盐酸添加量15 mL;精密度、重复性和稳定性试验结果的相对标准偏差(RSD)均小于2%,加标回收率94.60%~97.60%,RSD值均小于2%。该方法准确、可靠、重现性好,可用于黄水中总糖含量的测定。The determination of total sugar content in yellow water by 3,5- dinitrosalicylic acid (DNS) method had been developed. The optimum hydrolysis conditions of total sugar were summed up as follows: hydrolysis time was 30 min, hydrochloric acid concentration was 12 mol/L, and the addition level of hydrochloric acid was 15 mL. The optimum detection conditions were summed up as follows: DNS amount was 5 mL, coloration time was 7 min, and reaction temperature was at 90 ℃. The relative standard deviation (RSD) of precision, repeatability and stability were less than 2 %, the adding standard recovery rate were 94.60 % to 97.60 %, and RSD was below 2 %. This method was reliable, accurate and reproducible, and it could be used for the determination of total sugar content in yellow water.
