麸皮固体培养产朊假丝酵母菌的培养基优化研究

摘  要:选用产朊假丝酵母菌进行固体发酵,并通过单因素试验研究结果(接种量、料液比、发酵时间)确定产朊假丝酵母菌的最适宜生长条件。正交试验结果表明:固体培养基(包括麸皮、玉米粉)为底料,加入0.5%硫酸铵,0.05%磷酸二氢钾,不加硫酸镁,6%葡萄糖,加入适量蒸馏水(料液比为1∶1),接入12%产朊假丝酵母菌液,30℃培养36 h的条件下,产朊假丝酵母固体发酵培养基能得到较高的生物量。在该环境下产朊假丝酵母能够以最佳的生长状态生长。The solid fermentation was carried out by using Candida utilis, and the optimum growth conditions of Candida utilis were determined by single factor test results(inoculation amount, ratio of material to liquid and fermentation time). The orthogonal test results show that by using solid medium(including bran and corn meal) as the substrate, adding 0.5% ammonium sulfate, 0.05% potassium dihydrogen phosphate and 6% glucose, without adding magnesium sulfate, adding appropriate amount of distilled water (material to liquid ratio:1﹕1) and adding 12% bacteria liquid of Candida utilis, the solid fermentation medium of Candida utilis can obtain higher biomass under the condition of 30℃ for 36 h. In this environment, the Candida utilis can grow in the optimal growth state.
