摘 要:以青钱柳为材料,通过热水浸提,大孔树脂柱层析分离纯化,制备得到纯度大于90%的青钱柳黄酮和三萜。继而采用体外厌氧粪样混合培养与荧光原位杂交技术,评价青钱柳黄酮和三萜对于人体肠道菌群的调节作用。结果表明青钱柳黄酮和三萜类物质能有效促进肠道中有益细菌(双歧杆菌和乳酸菌)的增殖,而对于梭状菌和拟杆菌则有抑制作用,同时未显著影响肠道总菌群的数量。研究表明,青钱柳中的黄酮和三萜类物质,对于改善人体肠道环境、维护人体健康具有重要作用。In the present study,high purity(>90%)of flavonoids and triterpenoids were prepared from Cyclocarya paliurus by extraction with hot water and simple purification procedures of polyamide and AB-8 column chromatography.Then the modulatory effect of flavonoids and triterpenoids on human intestinal microbiota was evaluated through anaerobic mixed culture of fecal samples,and the bacterial populations were determined by fluorescent in situ hybridization(FISH).It can see that Cyclocarya paliurus flavonoids and triterpenoids promoted the growth of the beneficial bacteria(Bifidobacterium spp.and Lactobacillus/Enterococcus spp.),showed the inhibitory effect on Bacteroids-Prevotella spp.and Clostridium histolyticum group,while they did not affect the population of total bacteria.It can be inferred that Cyclocarya paliurus flavonoids and triterpenoids have certain regulation effects on intestinal environment by the modulation of the intestinal bacterial populations,contributing to the improvements of host health.
