摘 要:为了研究不同类别食品中不同形态叶黄素的分析方法,针对不同样品基质和叶黄素形态,分别选用超声波辅助提取法、酶解—超声波提取法和碱皂化—液液萃取法,建立并优化了测定叶黄素的高效液相色谱法。采用C18柱(4.6mm×250mm,5μm)进行检测,以甲醇—水为流动相进行等度洗脱,流速为1.0mL/min,柱温为35℃,检测波长为446nm,叶黄素在0.125~10.0mg/L范围内呈良好线性,相关系数为0.99991。不同类别食品的加标回收率为90.0%~102.2%,方法检出限为0.10mg/kg。采用该方法对各类食品样品中叶黄素的含量进行了测定,实验结果表明该方法适用范围广、针对性强、准确度高和重现性好,适用于各类食品中叶黄素的检测。In order to study the method for the determination of different forms lutein in various foods,different pretreatment methods were selected according to sample matrix and forms of lutein,including ultrasound-assisted extraction method,enzymatic hydrolysis-ultrasonic method and saponification-extraction method.A method for the determination of lutein by high performance liquid chromatography was established and optimized. HPLC separation of lutein was conducted on a C 18 column (4.6 mm×250 mm,5 μm ) with the mobile phase of methanol and water at the flow rate of 1.0 mL/min and the column temperature was 35 ℃. The detection was carried out at a wavelength of 446 nm.The linear range of the method for lutein were 0.125~10.0 mg/L with correlation coefficients not less than 0.999 91 .The recoveries of the lutein in various foods were from 90.0 % to 102.2 %.The detection limit of the lutein was 0.10 mg/kg.This method was applied in the investigation of lutein in various foods.The method has wide applicability,pertinence,good accuracy and reproducibility and is suitable for the detection of lutein in various foods.
